The experiment was conducted at the Federal Technological University of Paraná, Campus Dois Vizinhos in animal metabolism UNEPE, from June to August 2014. Four male sheep with ruminal cannula were used with 50 kg body weight . These animals were kept in cages with water fountains and individual feeders fed 20% of Tifton 85 hay and 80% concentrate. The treatments were 0, 2, 4 and 6% added protected fat from palm oil. The experimental period was 80 days, with 14 days of adaptation and a day of collection of ruminal fluid, which was used for the quantification of protozoa and pH determination. The experimental design was Latin square (4x4), with four levels of inclusion of fat and four periods. The pH was determined with a digital pH meter, at 0, 2, 4, 6 and 8 hours after feeding the diet. The collection to quantify the protozoa was taken at zero and two hours. It was used adapted Counting chamber Sedgewick-Rafter, the collected samples were fixed in formalin 18.5% and then applied lugol solution (100 mL of distilled water five and ten grams of iodine g of potassium iodide), the mean scores were considered independent made of 100 fields performed in duplicate analyzed under a microscope with 10X magnification. The feed supplied to the animals were samples taken from days of collection and made pre-drying in forced ventilation oven at 55 ° C for 72 hours for further chemical analysis of dry matter, organic matter, crude protein and neutral detergent fiber. Data were subjected to analysis of variance and the effect of the inclusion level of fat was analyzed by regression. No significant difference was observed in total count of ciliate protozoa the rumen of sheep (P> 0.05) for inclusion levels of dietary fat protected (0, 2, 4 and 6%), however significant differences (P <0.05) between times 0 and 2 hours after feeding. Regarding the pH, there was significant difference (P <0.05) compared the diets provided and the time of collection and showed minimum level 6.1 with 0.18% inclusion of fat, 5.35 hours after power. The use of protected fat of palm oil up to 6% in the concentrate in the diet of sheep does not affect the population of rumen protozoa. The adjusted number of protozoa chamber allows to count according to the methodology recommended in the literature. There is a reduction Protozoa with decreasing pH and constant presence of oxygen. O presente trabalho tem por objetivo quantificar os protozoários ciliados no rúmen de ovinos em relação à suplementação com gordura protegida de óleo de palma. O experimento foi realizado na Universidade Tecnológica Federal do Paraná, Campus Dois Vizinhos, na UNEPE de metabolismo animal, no período de junho a agosto de 2014. Foram utilizados quatro ovinos machos fistulados no rúmen com peso vivo médio de 50 kg. Estes animais foram mantidos em gaiolas com bebedouros e comedouros individuais alimentados com 20% de feno de Tifton 85 e 80% de concentrado. Nos tratamentos foram adicionados 0, 2, 4 e 6% de gordura protegida de óleo de palma. O período experimental foi de 80 dias, com 14 dias de adaptação e um dia de coleta do líquido ruminal, o qual foi utilizado para a quantificação dos protozoários e determinação do pH. O delineamento experimental utilizado foi Quadrado Latino (4x4), com quatro níveis de inclusão da gordura e quatro períodos. O pH foi determinado com pHmetro digital, nos tempos 0, 2, 4, 6 e 8 horas após o fornecimento da dieta. A coleta para quantificação dos protozoários foi feita nos tempos zero e duas horas. Foi utilizada Câmara de contagem adaptada de Sedgewick-Rafter, as amostras coletadas foram fixadas em formalina 18,5% e depois aplicadas em solução de lugol (100 mL de água destilada, cinco gramas de iodo e dez gramas de iodeto de potássio), foram consideradas as médias das contagens feitas em 100 campos independentes, realizadas em duplicata e analisados em microscópio com aumento de 10x. Dos alimentos fornecidos aos animais foram retiradas amostras dos dias da coleta e feita a pré-secagem em estufa de ventilação forçada a 55ºC por 72h para posteriores análises bromatológicas de matéria seca, matéria orgânica, proteína bruta e fibra em detergente neutro. Os dados foram submetidos a análise de variância e o efeito do nível de inclusão da gordura foi analisado por regressão. Não foi observada diferença significativa na contagem total de protozoários ciliados o rúmen de ovinos (P>0,05) para os níveis de inclusão de gordura protegida na dieta (0, 2, 4 e 6%), no entanto houve diferença (P<0,05) entre os tempos 0 e 2 horas após a alimentação. Em relação ao pH, houve diferença significativa (P<0,05) em relação as dietas fornecidas e o tempo de coleta e apresentou valor mínimo 6,1 com nível de 0,18% de inclusão de gordura, as 5,35 horas após a alimentação. O uso da gordura protegida de óleo de palma até 6% no concentrado em dieta de ovinos não afeta a população de ciliados no rúmen. A câmara adaptada de contagem de protozoários permite a contagem de acordo com a metodologia recomendada da literatura.
