Resumo: Apple culture is of great importance in the world scenario of temperate fruit production and its productivity is directly linked to the dormancy process. The present study aims to evaluate the mean time of budburst of three apple trees cultivars of low and medium chilling requirements and to test the efficiency of different protocols for RNA extraction in wood. Plant material were collected in a commercial orchard in Palmas, Southwest of Paraná. Cultivars used were “Eva”, “Fuji Suprema” and “Galaxy”. The evaluation of dormancy dynamics was done by single-node cutting methodology through the quantification of Mean Time of Budburst (MTB). The RNA extraction was done by the methods described by Salzman (1999), Zeng and Yang (2002), Quick Zol® and PureLink™ Plant RNA Reagent. The plants exposed to mild winters did not have a well-defined peak of MTB, and in face of the numerous protocols for nucleic acid extraction, efficiency, time, cost and residue should be tested.
