Resumo: The Neotropical region is considered unique among other terrestrial ecosystems, harboring a high diversity of taxa, including fish. Among the continental fish families present in the region, the Erythrinidaefamily stands out for its endemism in South and Central America, with a wide distribution in Brazilian basins, and being represented by three genera, among them HopliasGill (1903). The genus Hopliasencompasses two large groups, H. malabaricusBloch, 1794 and H. lacerdaeMiranda, 1908. H. malabaricusincludes several species, such as: H. malabaricus, H. mbigua, H. missioneira, among others present in the area of Upper Paraná. In addition to constituting a complex of species, with the presence of 7 karyomorphs, identified by the distinct diploid number. Classical (Giemsa staining and C-banding) and molecular (physical mapping of repetitive ribosomal sequences by fluorescent in situ hybridization -FISH) cytogenetic analyzes were performed in order to identify possible karyomorphs and the distribution pattern of these repetitive DNAs, present in the area of influence of Lake Itaipu in the municipality of Santa Helena-PR. Sixteen specimens were analyzed, 6 approved 2n=40 chromosomes (14m + 26sm, NF = 80) and 7 approved specimens 2n=42 chromosomes (22m + 20sm, NF = 84) thus characterizing as belonging to karyomorphs C and A , respectively . Thus, the three best (quality/quantity of metaphases) analyzed specimens were selected, with 2 individuals characterized by the karyomorph C (1 male and 1 female) and 1 individual of the karyomorph A (male). From the C-Banding, heterochromatic regions present in the pericentromeric region were evidenced in most chromosomes, regardless of the cytotype.FISH with 5S rDNA and 18S rDNA probes was performed in the 3 best individuals, and showed conversion between the diagnosed specimens. The absence of bitlomeric marking in one of the chromosomes of pair 13, only in the female of karyomorph C, stands out, thus being able to indicate a polymorphism, regardless of sex. For classic chromosomal analyses: the information advanced in this study is similar to that approved in other literature. And reinforce the presence of cytotypes A and C in the region of Lake Itaipu
