However, obtaining cultivars with high grain yield and high industrial quality is one of the main challenges of breeding programs. The direct methods of evaluation of the elastic properties of the flour are quite expensive and require large quantities of seeds, and it is possible to carry out only the advanced stages of the breeding programs. Another relevant factor is the possibility of analyzes that require small quantities of seeds, and that can be used in large scale in initial generations. Thus, the objective of this work was to validate the efficacy of analyzes that use a small volume of samples, for selection of superior wheat genotypes as industrial quality, in segregating populations of breeding programs. The results showed that the SDS-PAGE and SDS methods showed a high magnitude correlation with W, with coefficients of 0.56 and 0.47, respectively. Through SDS-PAGE analysis, 84.7% of the evaluated genotypes presented the 2 * allele, and 15.3% allele 1 for the GluA1 gene. In addition, 66.6% had the 7 + 9 allele, 20.5% the 13 + 16 allele, and 12.8% the 7 + 8 allele for the GluB1 gene. Finally, 77% of the genotypes presented the 5 + 10 allele, and 22% the 2 + 12 allele for the GluD1 gene. It is worth mentioning that 84.7% of the genotypes showed homogeneity regarding the protein profile of glutenins. With the use of the UMN marker19, 17.9% of the genotypes presented the 362bp fragment (Ax1 or -Null) and 82.1% the 344bp fragment (Ax2 *). It was also observed that, 20.51% of the genotypes presented the fragments 299pb (UMN25) and 415pb (UMN26), and 71,79% fragments 281pb (UMN25) and 499bp (UMN26). Thus, it was possible to observe that the methods of SDS, SDS-PAGE and use of the molecular markers, both requiring a small volume of samples, can be used to predict industrial quality in the early stages of breeding programs. 2020-11-16T20:38:01Z 2020-11-16T20:38:01Z 2018-06-20 bachelorThesis CONTE, Josiane.
