Resumo: The assessment is a diagnostic tool that lets it to analyze the different aspects involved in the process of teaching e learning. ENEM, on of the most assessments of graduation from High School in Brazil, was established in 1998 and since its inception has tried to take an innovate play compared to traditional assessments of access to higher education. Assuming that such assessment play the guiding role of high school, the present work aims to categorize and analyze the Biology questions in ENEM. Initially, the questions of tests applied from 1998 to 2012 were subject to content analysis and categorized by themes, and from 2009, also were analyzed the skills required. The data were statistically analyzed and it was observed that Biology is about of 20% of all tests. The most requested theme was The Interdependence of life, which includes the interactions between living beings and human intervention in the environment. In contrast, less appearance was the theme four – Transmission of life and mechanisms of genetic variability. Regarding skills developed, the more charged was to understand interactions between organisms and their environment. Based on the data and analysis, it was concluded that Biology has a significant number of questions in ENEM test and, in general, the questions present themselves well structured, requiring only increased uniformity with regard to themes and skills in each edition.
