The objective of this work was to develop and evaluate a viable and low cost method for reuse and maintenance of the strains. The study was carried out using strains from “Fermetis”, a national manufacturer, where beer was produced on a pilot scale, aiming at imitating the industrial process and later re-using post-fermentation yeasts. The yeasts were washed and isolated at a stage which contained only living and healthy yeasts. The strains preserved in the refrigerator started from a viability between 86 and 91% and after six months the viability decreased to about 60 to 65%, except for the US-05 yeast that better withstand the cold conditions, maintaining its viability in 76,5% for the present study. The freezing was not very efficient since most of the yeasts did not withstand the severe conditions. The reason behind this is that glycerol was added to the yeast in its stationary phase and the alcohol present in the solution became a cryosensitising agent. Thus, based on the results of this experiment, brewers and microbreweries will be able to maintain and reuse yeasts with a simple method that represents a large economy in brewer industry. A levedura é um ingrediente fundamental para a produção de cerveja e o seu reaproveitamento dentro da indústria cervejeira é de extrema importância. O objetivo presente trabalho foi desenvolver e avaliar um método viável e de baixo custo para o reaproveitamento e manutenção das cepas. O estudo foi realizado utilizando cepas da fabricante Fermetis onde cervejas foram produzidas em escala piloto, visando imitar o processo industrial para posteriormente reaproveitar as leveduras da pós-fermentação. As leveduras foram lavadas e isoladas em uma fase em que continha somente leveduras vivas e saudáveis. As mesmas foram armazenadas em temperaturas de geladeira, cerca 2 a 4ºC e outra parte foi congelada com uma solução crioprotetora de glicerol e ácido ascórbico em uma temperatura de -20ºC. As cepas preservadas em geladeira partiram de uma viabilidade entre 86 e 91% e depois de seis meses a viabilidade baixou para cerca de 60 a 65% com a exceção da levedura US-05 que resistiu melhor às condições de frio, mantendo sua viabilidade em 76,5% para o presente estudo. O congelamento por sua vez, não mostro-se muito eficiente, pois a maioria das leveduras não resistiu às condições severas. O motivo por traz disso é que a levedura foi adicionada ao glicerol em sua fase estacionária e o álcool presente na solução acabou se transformando em um agente criossensibilizador levando a baixa eficiência do método proposto.
